Microscope, Image capturing for Nikon Digital Insight DS-L2

The following materials and equipment associated with this procedure present exposure or physical health hazards. Safety precautions are prudent and mandatory.


Prior to performing this procedure, the following safety equipment must be accessible and ready for use: (e.g. chemical fume hood, biological safety cabinet, laminar flow hood, chemical spill kits)

Protective Equipment:

Prior to performing this procedure, the following personal protective equipment must be obtained and ready for use: (e.g. acid resistant gloves, safety eyewear, lab coat, chemical splash apron)

Waste Disposal:

This procedure will result in the follow regulated waste which must be disposed of in compliance with environmental regulations.


In the event that a hazardous material spills during this procedure, be prepared to execute the emergency procedure below.









Control Box
The DS-L2 control box:

  1. Please begin with following the SOP for the Nikon Optiphot microscope.
  2. Rotate the eyepiece on the microscope clockwise so that the image appears on the control box screen.

  3. Turn on camera control unit. The power button on the DS-L2 is located on the bottom of right side of the control box.
  4. To bring up the “ menu” , right click in the left quadrant of the screen.

  5. The exposure level, "Lev.",  should be green in color.

    Over exposure (red color), decrease the light source or adjust filters and diaphragms

    Under exposure (green color), increase the light source or adjust filters and diaphragms

  6. The first menu you will see is the CAM menu window. There is an "Adv1" (advance), "Adv2" and "REG." tabs. The CAM menu allows for the adjustment of camera operations and exposure for better picture quality. The option to switch between different forms of microscopy, DF FL (Darkfield/florescence), BF (Brightfield), DIC PH (Differential interferance contrast/phase contrast) are available at the bottom of this menu.

  7. Next, the “TOOL” menu window shows various different ways to measure two-point distances.

    For measuring options, click on the "T. Bar" button. This option gives the ability to take different measurements. T Bar

    1. Select your scale based upon the objective lens currently in use.
    • m1: 5X
    • m2: 10X
    • m3: 40X
    • m4: 100X (these lenses are kept in 2224C, due to their limited availability)
  8. Scale bars are also chosen and turned on here, click on the "Scale" button.

  10. To capture and save an image to the compact flash card:
    1. Go to “REC” menu window
    2. Choose what drive the images will be saved to. C: is the CF card. It is possible to attach a USB thumb drive to the side of the control box.
    3. Left click on the letters next to the "Dir.:"
    4. Type name for your folder.

    5. If desired, edit the file names by highlighting the box next to the word “File” in this menu, clicking again to reveal the alpha numeric pad and typing a new file name using the mouse.  “Ent” must be clicked to leave this screen whether you input anything or not.  “CL” clears all typing, “BS” erases the last character, and “SFT” changes between upper and lower case.

    6. If batch name change of the files is desired, in the "SETUP" menu window, under the "File Settings", the "File Name:" box can be changed using the numbers/letters located at the bottom of the screen. (similar to the DS-L1)
    7. It is possible to save either 1 image at a time or continuous images (1 every 2 to 30 seconds).Choose either "1-shot" or "Continue".
  11. To capture:
    • Select “C” on the “CAM” menu
    • Push “Capture” button on low right hand corner of the control unit
    • Right click mouse at any time to reveal a tool bar in lower right hand corner of the LCD screen and selecting the “CF” button
    • Select the "Save" button in the “REC” Screen menu.
  12. You can also view and download live or CF card saved images from any computer:
    1. Log onto any computer and open the web browser program.
    2. Make sure that the camera control unit is turned on.
    3. Go to the web address http://nomarski.engin.umich.edu
    4. Choose Room 1 to view capture and save live images.
    5. Choose room 2 to view images stored on the CF card.
    6. By this method you can save any image that it is being viewed, but you cannot transfer folders and groups of images.
  13. To transfer images:
    1. Remove CF card from control unit
    2. Obtain CF Card reader from 2224C or 2224A, they are not permitted to be taken out of the lab.
    3. Log onto a lab computer
    4. Insert card reader into USB port
    5. Transfer files from CF Card and delete pictures on CF card.
    6. Remove card reader from computer and return to 2224C or 2224A.
    7. Re-insert CF card into control unit
  14. You can also access camera control unit and compact flash storage by connecting laptop or other computer directly via a USB cable.
  15. Expect any images remaining on CF card to be deleted at any point (especially if card becomes full). Make sure to download images immediately following image capture.

  16. Before or after capturing your images, it is possible to view them by going to the "View" menu window.
